Abstract
Genotypic analysis in Vochysia guatemalensis (Vochysiaceae) by means of microsatellites. In this investigation, a clonal collection of 33 genotypes of Vochysia guatemalensis was analyzed utilizing microsatellites (SSR). Gene markers were obtained, adapted and validated, from a group of existing microsatellites for V. ferruginea. Fingerprinting analysis was performed in 33 clones, based on loci A1-20 and A1-15. Genotypes were also grouped based on their genetic distance. Among the four analyzed loci, the greater polymorphism was registered in loci A1-20 and A1-15, with seven alleles each one. In loci A1-5 and A1-35, five and six alleles were detected respectively. Polymorphic Information Quantity (PIQ) average was 0.77. Besides, the probability of finding two individuals random select in V. guatemalensis population with the same genotype (for A and C loci) were 8.34%; in other words, both loci have a 91.66% discrimination probability. Furthermore, occurrence probability for the more probable genotype (A3A3C1C1) of the total registered genotypes with two loci were 1.07%. For the clones grups with identical genotype, the 19 family with the A3A3C1C3 genotype showed an occurrence probability of 0.47%. Genetic diversity average was 4.72. Mean genetic distance among all genotypes in relation to the whole population was 68%. Two pairs of primers, out of five developed for V. ferruginea, gave useful information in V.guatemalensis to distinguish and assess the fingerprinting in 33 genotypes. Therefore, loci A1-20 and A1-15 are highly polymorphic in this V. guatemalensis collection, and may be useful in genetic certification processes, as well as in assisting gene conservation and breeding programs.
